Aberrant AHRR, ADAMTS2 and FAM184 DNA Methylation: Candidate Biomarkers in the Oral Rinse of Heavy Smokers

چکیده

Objective. To identify DNA methylation patterns of heavy smokers in oral rinse samples. Methods. Genome-wide data was imported from Gene Expression Omnibus GSE70977 using the GEOquery package. Two independent sets were analyzed: (a) 71 epigenomes cancer-free subjects (heavy n = 37 vs. non-smokers 31); for concordance assessment (b) 139 oral-cancer patients’ 92 47). Differential CpG positions and at regional level determined Limma DMRcate Bioconductor packages. The linear model included sex, age, alcohol consumption. statistical threshold set to p < 0.05. Functional gene prioritization analysis performed gene-targeted analysis. Results. In individuals without cancer smokers, FAM184B found with two differentially hypermethylated (p 0.012 after FDR adjustment), a region 48 bp an absolute difference >10% between groups 1.76 × 10−8). corresponding patients, we AHRR hypomethylated but also targeted analyses. Remarkably, ADAMTS2 diagnosis consecutive probes cg05575921 3.13 10−7) cg10208897 1.36 10−5). Conclusions: Differentially methylated AHRR, ADAMTS2, genes are biomarker candidates

Aberrant methylation of hypermethylated-in-cancer-1 and exocyclic DNA adducts in tobacco smokers.

Tobacco smoke has been shown to produce both DNA damage and epigenetic alterations. However, the potential role of DNA damage in generating epigenetic changes is largely underinvestigated in human studies. We examined the effects of smoking on the levels of DNA methylation in genes for tumor protein p53, cyclin-dependent kinase inhibitor2A, hypermethylated-in-cancer-1 (HIC1), interleukin-6, Lon...

Aberrant DNA Methylation in Keratoacanthoma

BACKGROUND Keratoacanthoma (KA) is a self-limiting epidermal tumor for which histopathological examination sometimes suggests malignancy. Based on inconsistent clinical views, KA can be regarded as both a benign tumor and a variant of squamous cell carcinoma (SCC). Aberrant DNA methylation frequently occurs in malignant tumors but it scarcely occurs in benign tumors. Whether aberrant methylatio...

Aberrant DNA Methylation in Cholangiocarcinoma

Cholangiocarcinoma is an epithelial malignancy arising in the region between the intrahepatic bile ducts and the ampulla of Vater at the distal end of the common bile duct. The effect of current chemotherapy regimens against cholangiocarcinoma is limited, and the prognosis of patients with cholangiocarcinoma is poor. Aberrant DNA methylation and histone modification induce silencing of tumor su...

DNA Methylation and Its Role in the Development of Leukemia

Epigenetic changes play an essential role in cancer pathogenesis. It has been established by next-generation sequencing that more than 50% of the human cancers carry mutations in mechanisms involved in the organization of the chromatin and epigenetic regulations. DNA methylation is among the most common epigenetic changes in leukemia. In contrast to DNA mutations which are passively inherited f...

the analysis of the role of the speech acts theory in translating and dubbing hollywood films
